MOF-mediated PRDX1 acetylation regulates LPS-induced macrophage activation

چکیده

Abstract Macrophages respond to environmental cues and play a crucial role in host defense, inflammation, tissue homeostasis. Macrophage activation undergoes metabolic transcriptomic reprogramming adapt tailor an appropriate response, regulating innate immunity. Reactive oxygen species (ROS) have been shown pivotal macrophage activation. Nevertheless, the detailed molecular mechanism of how redox signals direct is not fully elucidated. Here, we uncover for lysine acetyltransferase MOF (also known as KAT8) inflammatory via modulating acetylation peroxiredoxin 1 (PRDX1), H 2O 2scavenger. We demonstrate that PRDX1 novel substrate MOF. dynamic reversely regulated by HDAC6 SIRT2. fine-tunes its peroxidase activity, leading modulation intracellular ROS level upon LPS stimulation. Moreover, appears participate LPS-induced regulation 2-mediated signal transduction.

Ginger extract inhibits LPS induced macrophage activation and function

BACKGROUND Macrophages play a dual role in host defence. They act as the first line of defence by mounting an inflammatory response to antigen exposure and also act as antigen presenting cells and initiate the adaptive immune response. They are also the primary infiltrating cells at the site of inflammation. Inhibition of macrophage activation is one of the possible approaches towards modulatin...

SCRG1 suppresses LPS-induced CCL22 production through ERK1/2 activation in mouse macrophage Raw264.7 cells

Recently, we identified the scrapie responsive gene 1 (SCRG1) secreted from mesenchymal stem cells (MSCs) and its receptor bone marrow stromal cell antigen 1 (BST1) as positive regulators of stem cell qualities such as self‑renewal, migration abilities, and osteogenic differentiation potential. Here, we examined the effect of the paracrine activity of SCRG1 in macrophages. The mouse macrophage‑...

The LPS-induced neutrophil recruitment into rat air pouches is mediated by TNFα: likely macrophage origin

The role of resident cells during the lipopolysaccharide (LPS)-induced neutrophil recruitment into rat air pouches was investigated. In this model, LPS (Escherichia coli, O55: B5 strain; 2-2000 ng) induced a dose- and time-dependent neutrophil recruitment accompanied by the generation of a tumour necrosis factor-alpha (TNFalpha)-like activity. Dexamethasone (0.05-5 mug) and cycloheximide (6 ng)...

Silibinin Inhibits LPS-Induced Macrophage Activation by Blocking p38 MAPK in RAW 264.7 Cells

We demonstrate herein that silibinin, a polyphenolic flavonoid compound isolated from milk thistle (Silybum marianum), inhibits LPS-induced activation of macrophages and production of nitric oxide (NO) in RAW 264.7 cells. Western blot analysis showed silibinin inhibits iNOS gene expression. RT-PCR showed that silibinin inhibits iNOS, TNF-α, and IL1β. We also showed that silibinin strongly inhib...

-Mediated Activation of STAT1 a Regulates Growth Factor–Induced Mitogenesis

Immunomodulatory cytokines and growth factors act in a complex network to regulate diverse biologic processes. Pretreatment of two types of human vascular pericytes, liver fat–storing cells or glomerular mesangial cells, with IFNg dramatically enhanced DNA synthesis in response to PDGF or EGF. IFNg by itself had very little effect on DNA synthesis. At least 24-h exposure of the cells to IFNg is...
